چکیده

The indole-J-glycerol phosphate synthetase of Escherichia coli was purified to apparent homogeneity and was crystallized. The enzyme is a single polypeptide chain of 45,000 molecular weight with 1 NHz-terminal methionine residue. The amino acid composition of the protein was determined. The rate of synthesis of this polypeptide chain in E. coli was found to be identical with the rate of synthesis of the polypeptide chains of the tryptophan synthetase a! and p2 subunits.

Mechanism of 3-methylanthranilic acid derepression of the tryptophan operon in Escherichia coli.

3-Methylanthranilic acid (3MA) inhibits growth and causes derepression of the tryptophan biosynthetic enzymes in wild-type strains of Escherichia coli. Previous reports attributed this effect to an inhibition of the conversion of 1-(o-carboxyphenylamino)-1-deoxyribulose 5-phosphate to indole-3-glycerol phosphate and a consequent reduction in the concentration of endogenous tryptophan. Our studi...

Mutants of Escherichia Coli Defective in the B Protein of Tryptophan Synthetase. Ii. Intragenic Position.

investigation of the genetic control of the structure of a specific protein likely to be influenced by the method of selecting the mutants used for study. If the protein has enzymatic activity and use is made of the lack of enzymatic activity in mutants for identification and selection, there will doubtless be many alterations of the protein which pass unnoticed because their effect on the enzy...

A Novel Tryptophan Synthase -Subunit from the Hyperthermophile Thermotoga maritima

Tryptophan synthase catalyzes the last two steps in the biosynthesis of the amino acid tryptophan. The enzyme is an complex in mesophilic microorganisms. The -subunit (TrpA) catalyzes the cleavage of indoleglycerol phosphate to glyceraldehyde 3-phosphate and indole, which is channeled to the active site of the associated -subunit (TrpB1), where it reacts with serine to yield tryptophan. The Trp...

Two mechanisms of allelic complementation among tryptophan synthetase mutants of Saccharomyces cerevisiae.

Two different types of allelic complementation were observed in tryptophan synthetase mutants of the yeast Saccharomyces cerevisiae. Each type is associated with a different mechanism for the enzymatic conversion of indole-3-glycerol phosphate (InGP) to tryptophan. Mechanism I is utilized by a hybrid tryptophan synthetase that resembles, but is not identical with, the wild-type enzyme. Mechanis...
